I will go down with this ship.
If you do not believe in Destiel, you have to at least agree that this is a very complex relationship. Perhaps Castiel is just in love with Dean in the sense that Dean gives him a brotherly love that Cas has never felt from the other angels. Perhaps Dean is just in love with Cas in the sense that Cas is the first angel who has reached out to him. Besides Sammy, he has Cas. Cas is who he's always been wanting in his life.
But I'm sorry, there is no way that Destiel wont be canon. I mean, have you seen any episode involving the two of them?
Let this lone fact sink in: every long look of admiration, every stare, and every suggestive conversation was planned by the directors. The writers of Supernatural specifically wrote in those moments. There was at least one time where directors said "Cut! Jensen, we need you to stare at Misha for a little longer. Thanks. And, action!"
I have already proved how Dean is bisexual, and this is my list of evidence that suggests that he's in love with Castiel.

Dean is only used to familial love, because he has never really had a successful relationship (It's ironic because Cas is not used to familial love). Dean couldn't exactly love Lisa, romantically anyway. I think it means that Dean hasn't really recognized the state of being in love, so it's only natural that he's the only person who doesn't see the chemistry between he and Cas.
Balthazar: "I think you have me confused with the other angel. You know, the one in the dirty trench coat who's in love with you."
Meg: "Go ask him, he was your boyfriend first."
Uriel: "You see, he has this weakness. He likes you."

When Cas confesses that he had sex with April, Dean chokes on his drink. Not Sam, Dean. Even that is slightly significant.

Dean didn't believe in angels. He didn't believe that there was something/someone out there that could make miracles happen; that could make lives better. Dean has lived with a lot of guilt, sadness, disappointment, low self-worth, anger, pain (physical and emotional)... shall i go on? I don't blame Dean for not believing in angels, could you? For all of the years he has felt those horrible emotions... no one, not even a freaking angel, has tried to help. Until Cas. Cas gave Dean a reason to believe in the heavenly side of things. Granted, angels are dicks... but not Cas.
That belief is called faith, which is pretty much love in the heart of an angel. Surely Cas has felt Dean's undeniable faith, because he gave up everything for Dean Winchester in return. Not Sam, not all of his fellow angels, not his Father... Dean. Cas betrayed his orders from Heaven. He made decisions out of his blind love for Dean, and chose him over everyone else. He raised Dean from Hell, he stood by this human through practically Hell-on-Earth. Dean either tries not to notice the infatuation, or just doesn't recognize it because Dean thinks that he doesn't deserve to be loved.

Dean kept Cas' coat while he was off being God and internally combusting, with hope that Cas would return.

So, there's a scene in the episode where Dean, Sam, and Cas track down Cupid. Dean goes to get a drink at a bar, and he seems to be watching some hunting show where the hunter is aiming a bow. Okay, a bit coincidental, don't you think? Then Cas approaches the bar to sit next to Dean, and that is what's extremely important.
